چکیده

HE mutant alleles of the dumpy locus produce a number of phenotypic Teffects, viz., an obliquity of the wings ( 0 ) , a lethal effect (1) , and hypodermal irregularities of the thorax called vortices (v) . The genetics of the dumpy locus has been described by CARLSON (1959). The alleles may express any one of the three effects singly (dp", d p z , dp") or in any combination with one another (dpoz , dpzv , dp"", d p o z v ) . There is also a comma mutant ( c m ) characterized by commalike invaginations at the anterior thorax. There is some intragenic complementation among the alleles in the trans configuration, for example, ou/Z is wild' type. Heterozygous combinations manifest the trait common to both alleles, for example, ou/u manifests thoracic abnormalities. This paper is about allelic combinations which produce thoracic abnormalities. A total of five thoracic abnormalities have been described. The existence of three of these, viz., first vortex (v), second vortex (2nd v) and comma (cm) has been known for some time. They are expressed on the scutum and each abnormality when manifested causes some local disturbance in the bristle pattern. The other two abnormalities, pre-episternal pit and posterior invagination, have only recently been described (METCALFE 1969). The presence of these abnormalities, owing to their positions on the thorax, is not associated with disturbances in the pattern of microchaetae. All five thoracic abnormalities have various forms; namely, pits, eruptions, or raised pits (an expression intermediate between the former two) of the cuticle. This paper reports the phenotypic patterns of abnormalities produced by the different v-producing alleles; the developmental history of the thoracic abnormalities which determine the variations in form of the abnormalities; and the morphogenetic distinctiveness of each v-producing allele. The results are presented in four sections: SECTION I describes the phenotypic patterns of abnormalities manifested by the v-producing allelic combinations. SECTION 11 reports the development of the thoracic abnormalities (invaginations of the hypodermis) ; their similarities and differences; and associated muscle breakdown in a dp"" stock-this genotype being chosen because it manifests four of the five abnormalities and is not lethal when homozygous. SECTION 111 deals with the internal as opposed to the external phenotype of adults of v-producing allelic combinations, i.e., differences in the size of invaginations and the amount

A Molecular Analysis of Mutations at the Complex dumpy Locus in Drosophila melanogaster

The Drosophila dumpy gene consists of seventy eight coding exons and encodes a huge extracellular matrix protein containing large numbers of epidermal growth factor-like (EGF) modules and a novel module called dumpy (DPY). A molecular analysis of forty five mutations in the dumpy gene of Drosophila melanogaster was carried out. Mutations in this gene affect three phenotypes: wing shape, thoraci...

Aconitase and Developmental EndPointsasEarly IndicatorsofCellularToxicity Induced by Xenobiotics in Drosophila Melanogaster

Background: In this study, the toxicity of the different xenobiotics was tested on the fruit fly Drosophila melanogaster model system.  Methods: Fly larvae were raised on food supplemented with xenobioticsat different concentrations (sodium nitroprusside (0.1-1.5 mM), S-nitrosoglutathione (0.5-4 mM), and potassium ferrocyanide (1 mM)). Emergence of flies, food intake by larvae, and pupation h...

The developmental genetics of apterous mutants of Drosophila melanogaster.

HE apterous (ap) mutant of Drosophila melanogaster was first found by MISS EDITH M. WALLACE in 1913 while working in the laboratory of DR. T. H. MORGAN. This mutant,'which occurred spontaneously, was shown by C. W. METZ (1914) to be recessive and located on the second chromosome near the black ( 6 ) locus. Subsequently, other apterous mutants were found, such as up4, which was localized at 2-55...

Mutagenesis at a complex locus in Drosophila with the monofunctional alkylating agent, ethyl methanesulfonate.

use of chemical mutagens in Drosophila usually results in the production T:mutants which are composed of mutant and nonmutant tissue. Mutants of this type are referred to as mosaic or fractional, as opposed to the complete or whole body mutants so commonly found in studies of X-ray mutagenesis. For the detection of mosaic and complete mutants, as well as for general mutagenesis studies, the mos...

Dumpy mutations following X-irradiation of Drosophila melanogaster mature sperm in oxygen or in nitrogen.

A comparison was made of the oxygen enhancement pattern among the different kinds of dumpy mutations (olv, ov, ol, lv, o and v types), yellow mutations on the scute-8 chromosome, white, miniature and forked mutations, and the marker losses in the doubly marked Y chromosome [BS Y sc8 (y+)], all of which were induced by X rays in mature sperm of Drosophila melanogaster. The results indicate that ...
